Some lectins in this group stimulate distinct T- and B- cell functions, such as Jacalin, which binds to the T-antigen and acts as an agglutinin. This domain is found in 1 to 6 copies in lectins. The domain is also found in the salt-stress induced protein from rice and an animal prostatic spermine-binding protein.

- Jacalin, a tetrameric two-chain lectin (66,000 Mr) from jackfruit seeds,is highly specific for the tumour associated T-antigenic disaccharide. Thecrystal structure of jacalin with methyl-alpha-D-galactose reveals thateach subunit has a three-fold symmetric beta-prism fold made up of threefour-stranded beta-sheets. The lectin exhibits a novelcarbohydrate-binding site involving the N terminus of the alpha-chainwhich is generated through a post-translational modification involvingproteolysis, the first known instance where such a modification has beenused to confer carbohydrate specificity. This new lectin fold may becharacteristic of the Moraceae plant family. The structure provides anexplanation for the relative affinities of the lectin for galactosederivatives and provides insights into the structural basis of itsT-antigen specificity.
