| Interpro abstract (IPR001754): | Orotidine 5'-phosphate decarboxylase (OMPdecase) [(PUBMED:2835631), (PUBMED:1730672)] catalyses the last step in the de novo biosynthesis of pyrimidines, the decarboxylation of OMP into UMP. In higher eukaryotes OMPdecase is part, with orotate phosphoribosyltransferase, of a bifunctional enzyme, while the prokaryotic and fungal OMPdecases are monofunctional protein. Some parts of the sequence of OMPdecase are well conserved across species. The best conserved region is located in the N-terminal half of OMPdecases and is centred around a lysine residue which is essential for the catalytic function of the enzyme. This entry also includes enzymes such as 3-hexulose-6-phosphate synthase EC 4.1.2.43 and 3-keto-L-gulonate-6-phosphate decarboxylase EC 4.1.1.85. |

The nucleotide sequence of the Myxococcus xanthusorotidine-5'-monophosphate decarboxylase (OMP DCase) gene was determined.The derived protein sequence is not closely related to other prokaryoticOMP DCase sequences; nor is it closely related to any eukaryotic OMP DCasesequences. Progressive multiple alignment of the M. xanthus OMP DCaseprotein sequence with 19 other OMP DCase sequences revealed four conservedregions present in all 20 sequences. Ten entirely conserved residues werefound in these four regions and one region contains a tight cluster of 5conserved residues, certain of which may be catalytically active residues.A second open reading frame was found upstream of uraA and oriented in thesame direction as uraA. A stretch of 21 consecutive pyrimidine (C or T)residues were found in the intercistronic region between the potentialribosome-binding site of uraA and the UGA stop codon of the upstream openreading frame. RNA directly upstream of the pyrimidine run, including theUGA stop codon of the upstream open reading frame, could be folded into astable hairpin structure resembling Rho-independent terminators ofEscherichia coli. Expression of the uraA gene may be regulated by anintercistronic transcription termination mechanism.

A Dictyostelium discoideum DNA fragment that complements the ura3 and theura5 mutants of Saccharomyces cerevisiae has been sequenced. It containsan open reading frame of 478 codons capable of encoding a polypeptide ofmolecular weight 52475. This gene, named DdPYR5-6, encodes a bifunctionalprotein composed of the orotate phosphoribosyl transferase (OPRTase) andthe orotidine-5'-phosphate decarboxylase (OMPdecase) domains described forUMP synthase in mammals. The existence of separate domains for the twoactivities was suspected because deletion of the N-terminal coding segmentof the gene eliminated the ura5 but not the ura3 complementing activity.We have now confirmed that the two parts of the open reading frame sharehomology with known OPRTase and OMPdecase sequences. Several blocks ofsequence are conserved among OPRTase from bacteria, fungi and slime moldand one of them corresponds to the consensus sequence forphosphoribosylbinding sites. The OMPdecase domain shows extensivesimilarity with the yeast and Neurospora crassa enzymes, suggesting thatthey have evolved from an ancestral gene which was fused to the OPRTasegene in D. discoideum. It is less related to the bacterial enzyme but allthese sequences present conserved blocks of homology which could identifythe active site. The codon usage is strongly biased in a manner similar tothat found for other D. discoideum genes. The flanking DNA containshomopolymers of A and T and alternating sequences that are characteristicof the gene organization in D. discoideum.
